Nathan 8. Kline, M.D.

Director of Research

Rockland State Hospital
Orangeburg, New York 10962
Dear Nate,
As you know, I have been most
interested in the development
of narcotic antagonists as a treatment ior opiate
dependence. After many
years of effort, the model has been supported by clinical studies, and
the qualities of an ideal antagonist have been defined. The National
Institute of Mental Healhh, the SAODAP of the White House, NACC
the
State of New York, and the City of New York are all interested inof these
programs. They are making research funds available, and their
support
has much to commend

it.

However, the principal agencies that have the facilities to
produce the necessary formulations, the industrial laboratories,
preoccupied by their prior committments, and some effort should bearemade
to bring the needs of this program to the attention of the formulating
chemists. With this in mind, I would ask that you consider approaching
some of your foundation friends, with the idea
a special award be
established for the production of a successful that
formulation with the

following characteristics:

specific narcotic antagonism, similar to naloxone;
duration on single administration of 90 days, with
antagonism to injected heroin, 25 mg/ch/2 minutes;
tolerated by man ('safety'); and,
without agonistic features.

is the most likely starting substance now at hand, but other
candidates may be l-cyclazocine, diprenorphine (M—SOSO),
other
specific antagonists now under study at Bristol, Geigy, or orother
labs.

Naloxone

A single prize of $50,000
or $100,000 for 36 months, well
publicized and supported by the authority of one of the leading
found~
ations may serve this program handsomely.

I hope you agree, and raise the question at the best levels.
Sincerely yours,
Max

Fink, M.D.
