Cole Superintendent Boston State Hospital Boston, Massachusetts 02124 Dear Don, In l96h665, in St. Louie, Turen and I studied the effects of thioridaaine (5 mg/kg), chlordiazepoxide (1 mg/kg), each alane; and thioridazine (2.5 mg/kg) and chlordiezepoxide (0.5 ng/kg) combined in a croseover design in 24 chronic schizophrenics. Dosages were maintained for 8 weeks and there was a placebo period of 8 weeks between each dosage. The clinical effects have been published, as anticipated. and the findings were In the EKG, BR, EP and hemogram data, we found consistent changes with thioridazine alone or combined, and not with chlordiezepoxide. In my hands, there are no differences in clinical activity between thioridazine, fluphenazine and butaperazdne. Since fluphenazine exists in 4 different formulations, has a very high therapeutic/safety margin (doses fro: 20 to 1200 mg have been given by us), and induces little or no EKG changes, I see no Justification for continuing the use of thioridazine (ace Ban, pg. 150). �,ww Dr. Cole June 29, 1971 ~2~ Similarly, I wonder about the very rapid approval by the FDA of meeoridazine, a sister compound, for which seemingly excessive claims have been made (in therapy of alcoholism and anxiety). So, my view for both compounds is tempered by the variety of antipsychotic drugs available, and the suggestion of greater effects on the EKG.
