umiyv-mwtrn,

Dear Dave,

In 1965, NIMH supported a collaborative study of schizophrenic
patients treated with placebo, high dose (1600 mg/day) and low dose
(400 mg/day) chlorpromazine for 6 months. In one study, by
H. C. B. Denber at Manhattan State Hospital, EEG records were collected
pre-drug and monthly, from 2 lead pairs (RQ~RT ear, RF-RO). The records
were accorded on magnetic tape and the tapes were analyzed in 1966 in
St. Louis on the IBM 1710 data analysis system. Sample size was 30
seconds; 320 samples per second; without prefiltering; period analysis
for baseline cross and first derivative.

changes were found in an increase in theta activity and a decrease in
frequencies above 40 cps. Variability of frequencies also decreased.

These two studies indicate that changes in the EEG after
chlorpromazine are drug specific, dose related in degree of change,
and persist for periods up to 6 months of continuous treatments.

It is clear

similar studies, that the EEG
changes with psychoactive drugs are not "side~effects" or “abnormalities"
but direct effects of the drugs, reflecting the central processes which
from

these,

and many

they are supposed to effect.
We

failed to

make

direct correlations between the

EEG

change and

behavioral improvement with the two doses of chlorpromazine in these
patients, although there was a difference of drug gs; placebo. The
differences between drugs were small,.and while significant in amount,
the behavior changes were small. For a detailed discussion of the
behavior changes, see Prien and Cole (Arch. gen. Psychiat., lg; 482-695,
1968).

If these comments are useful, they may be quoted either as "personal
communication" or as presented at the Society for Biological Psychiatry,
May 1967. Enclosed are abstracts and exhibits of the presentation.

It was a pleasure to discuss these interesting problems. I met
with Turan Itil in Aspen, and told him of your interest in the
schizophrenia study in Copenhagen. He is the principal investigator
and should write to you when he sets up the project.
