Dear Wilma,

Enclosed is our interim report and revised protocol in
our studies of GP-41299. Pages 1-8 are the same as the copies
which you have.
I have revised the clinical trial sections, following
your suggestions and after discussing the possibilities with

maintained on either chlordiazepoxide or doxepin for many
months, and also some schizoPhrenics who have been maintained
on phenothiazines. These patients can be placed on placebo.
In the event that symptoms recur, we will be able to begin with
GP-41299. We will have to determine the dosage first and if
an effective range is observed, I would then plan to treat
alternate patients with chlordiazepoxide or GP-41299.

Concurrently, I plan to select a few patients admitted
for severe anxiety or schizophrenia to our in—patient beds and
begin Open clinical trial. In these the possibility that they

Part two of the EEG trial (GP vs. placebo) will begin
within the next few weeks. The clinical studies can begin
early in November and for this I request a supply of 25 mg
capsules of GP-41299. If available, 1000. It would be helpful
to have 200 additional placebo capsules.

Ame.

While I would like to suggest that equivalent capsules
of chlordiazepoxide each containing 25 mg, I am not sure that
this is the best strategy. If it turns out that a clinical
effective dosage of GP 41299 is much higher than 150 mg per day,
then a comparison with 25 mg chlordiazepoxide would have a
limitation. Either the capsules could be made up now or we
might wait until we have some data to permit a better judgment.

If this preliminary protocol is satisfactory,
